In LNCaP Cells Inhibition of BCL-2 by Antisense Oligonucleonucleotides Results in Compensatory Changes in Apoptosis

چکیده
Antisense oligonucleotides (oligos) have been evaluated for treating prostate cancer in both in vivo and in vitro models. Although most oligos contain a single mRNA binding site, our laboratory evaluates bi-specific oligos directed towards two proteins. This study evaluates the growth inhibition in vitro of the LNCaP cell line employing monoand bispecific oligos directed against BCL-2 [the second binding site was directed against the epidermal growth factor receptor (EGFR)]. These oligos were administered with lipofectin as part of a nanoparticle delivery system. Additionally, g, the expression of five apoptosis regulatory proteins, BCL-2, bax, caspase-3, clusterin and AKT-1 was evaluated by RTPCR.

Effects of BCL-2 suppression by antisense oligonucleotides on additional regulators of apoptosis compensatory change in non-targeted protein expression.
Antisense oligonucleotides (oligos) have been employed against prostate cancer in both in vivo and in vivo models. While most oligos contain a single mRNA binding site, our laboratory has developed bispecifics directed towards two. Previous work has determined that when oligos are used to suppress the expression of individual proteins in highly regulated physiologic processes, additional protei...
متن کاملAndrogens induce resistance to bcl-2-mediated apoptosis in LNCaP prostate cancer cells.
We describe an in vitro model for prostate cancer treatment that suggests a potential benefit for combined androgen ablation and cytotoxic chemotherapy. Androgen treatment of the LNCaP hormone-dependent human prostate cancer cell line induces increased expression of the BCL-2 protein. Increased levels of this protein are known to mediate inhibition of apoptosis. LNCaP cells, however, did not un...
متن کاملProgression to androgen independence is delayed by adjuvant treatment with antisense Bcl-2 oligodeoxynucleotides after castration in the LNCaP prostate tumor model.
Bcl-2 has emerged as a critical regulator of apoptosis in a variety of cell systems and is up-regulated during progression to androgen independence in prostate cancer cells. The objectives of this study were to characterize changes in Bcl-2 after androgen withdrawal and during progression to androgen independence in the human prostate LNCaP tumor model and determine whether adjuvant use of anti...

Background and purpose: Telomerase activity has a major role in acute promyelocytic leukemia (APL). It also has a critical role in disease recurrence. This research aimed at studying the cytotoxic effects of telomerase inhibition using oligonucleotide-based molecule against human telomerase RNA template (hTERC antisense) and non-nucleoside small molecule targeting catalytic subunit (BIBR5132) o...
متن کاملSuppression of Bcl-2 by Antisense Oligonucleotides is Compensated through Increased Activity of the Androgen Receptor and Co-Activators but not 5-alpha Reductase
Antisense oligonucleotides (oligos) have targeted regulatory proteins in both in vivo and in vitro prostate cancer models. In efforts to identify compensatory changes in the expression of non-targeted genes this study evaluated monoand bispecific oligos capable of targeting and equally suppressing the expression of bcl-2 (an apoptosis inhibitor). Previous studies have shown that oligo treated L...
